    How an Essential Vitamin B-Derived Nutrient Concentrates in Mitochondria < Yale School of Medicine

    There’s a molecule that our body makes from vitamin B5 that is critical for all of the metabolic processes essential for human life. And when something goes wrong in that molecule’s production, it affects nearly every organ system in our body and causes a number of diseases.

    Researchers have discovered that up to 95% of this molecule—called essential cofactor coenzyme A (CoA)—is located inside mitochondria, organelles that supply cellular energy and regulate cellular metabolism. But what has not been clear is how CoA gets there.

    Reporting in Nature Metabolism, Yale researchers have now uncovered that CoA is trafficked into mitochondria and have identified the mechanisms responsible.

    This information, the researchers say, is important for future considerations about when and where to target treatments for diseases in which CoA is implicated.

    Mitochondria import CoA

    One reason it has been difficult to pin down how CoA gets to mitochondria is because CoA doesn’t exist by itself. As a cofactor, it attaches to lots of other molecules, and when bound together, the resulting molecules—called CoA conjugates—have entirely different structures.

    “That makes this difficult to study, to have a holistic understanding about CoA,” says senior author Hongying Shen, PhD, associate professor of cellular and molecular physiology at Yale School of Medicine and a member of the Systems Biology Institute at Yale West Campus.

    To address this challenge, Shen’s lab developed a new method to profile all of the different CoA conjugates using their expertise in mass spectrometry—a technique that can identify and quantify different molecules.

    With this new approach, the researchers were able to identify 33 different CoA conjugates in whole cells as well as 23 CoA conjugates in mitochondria.

    The question then was whether the CoA conjugates in the mitochondria were made there or brought in from elsewhere.

    In additional experiments, the researchers discovered that the enzyme required to make CoA largely exists outside of mitochondria. Further, when they made cells that lacked the molecular transporters that can move CoA around, mitochondria had far less CoA.

    “These findings strongly support the idea that CoA is being imported into mitochondria, and these transporters are required for that to happen,” says Shen.

    From function to disease

    This study advances the fundamental understanding of CoA and how it gets to where it needs to be in order to perform its essential functions. That, in turn, sheds light on how disruptions of this process might contribute to illness.

    For instance, mutations in the genes that produce CoA transporters are associated with diseases such as encephalopmyopathy, a disorder that can include neurodevelopmental delay, epilepsy, and decreased muscle tone. Mutations in the enzymes that produce CoA have been implicated in neurodegeneration.

    Going forward, Shen and her lab are investigating what role mitochondria CoA regulation plays in specialized cell types, such as neurons, and how dysregulation might contribute to disease.

    “In the context of brain disorders, such as neurodegeneration and psychiatric disorders, there’s an emerging idea that dysregulated mitochondrial metabolism is a contributor,” says Shen, who notes that her interest in micronutrients like vitamin B5 is part of a long Yale history in metabolism stretching back more than a century to Lafayette Mendel, PhD, former Sterling Professor of Physiological Chemistry whose discoveries included vitamin A and vitamin B complex in the mid-1910s.

    “We hope to contribute to this legacy and with our deep understanding of cellular metabolism, we hope we can provide new directions for diagnosing and possibly treating these diseases down the road.”

    FDA Grants Premarket Approval to Idylla CDx MSI Test for MSI-H CRC

    The FDA has granted premarket approval to the fully automated, cartridge-based, “sample-to-result” Idylla CDx MSI Test, a companion diagnostic test for identifying eligible patients with microsatellite instability–high (MSI-H) colorectal cancer (CRC) who may benefit from treatment with nivolumab (Opdivo) as monotherapy or in combination with ipilimumab (Yervoy).1

    “The approval of this new MSI companion diagnostic for patients with CRC is a meaningful achievement from our collaboration with Biocartis and a strong reflection of our Precision Medicine strategy at Bristol Myers Squibb,” Sarah Hersey, vice president of Precision Medicine, Bioanalytical and Translational Sciences at Bristol Myers Squibb, stated in a news release. “Rapid and accurate diagnosis is crucial to enabling access to appropriate therapeutic approaches, and this latest advancement exemplifies our commitment to delivering innovative, targeted solutions that have the potential to improve outcomes for patients.”

    The Idylla CDx MSI Test was designed to be used on the Idylla Platform. This qualitative test can detect a panel of 7 monomorphic biomarkers (ACVR2A, BTBD7, DIDO1, MRE11, RYR3, SEC31A, and SULF2) that are indicative of MSI in samples of CRC tissue. This test can be run with less than 3 minutes of hands-on time and can deliver results in less than 3 hours.

    What Was the Design of CheckMate 8HW?

    The multicenter, open-label trial randomly assigned patients with immunotherapy-naive unresectable or metastatic CRC with known MSI-H or mismatch repair–deficient (dMMR) status in a 2:2:1 fashion to receive treatment with nivolumab monotherapy (n = 353), nivolumab plus ipilimumab (n = 354), or investigator’s choice of chemotherapy (n = 132).2 In the first-line setting, the trial compared nivolumab plus ipilimumab vs chemotherapy. Across all lines of therapy, the trial compared nivolumab plus ipilimumab vs nivolumab monotherapy.

    Where Does Nivolumab Currently Fit Into the MSI-H CRC Treatment Paradigm?

    Notably, nivolumab plus ipilimumab was FDA approved on April 8, 2025, for the treatment of adult and pediatric patients 12 years of age or older who have dMMR or MSI-H unresectable or metastatic CRC.3 On the same date, the FDA also converted the accelerated approval of nivolumab monotherapy to a regular approval for the treatment of adult and pediatric patients 12 years of age or older who have MSI-H or dMMR metastatic CRC and have progressed following treatment with fluoropyrimidine, oxaliplatin, and irinotecan.
    The efficacy of nivolumab was assessed in CheckMate 8HW, which showed that patients with dMMR/MSI-H metastatic CRC who received first-line nivolumab plus ipilimumab achieved a median progression-free survival (PFS) that was not reached (NR; 95% CI, 38.4 months-not estimable [NE]) vs 5.8 months (95% CI, 4.4-7.8) among those who received chemotherapy (HR, 0.21; 95% CI, 0.14-0.32; P < .0001). Additionally, across all lines of therapy, the median PFS was NR (95% CI, 53.8 months-NE) with nivolumab plus ipilimumab vs 39.3 months (95% CI, 22.1-NE) with nivolumab monotherapy (HR, 0.62; 95% CI, 0.48-0.81; P = .0003).

    Man ‘stole Banksy for his love hearts art collection’ court told

    A builder accused of stealing a Banksy print has told a court he wanted to add it to his existing collection of artwork featuring love hearts.

    Girl with Balloon, valued at £270,000, was taken from the Grove Gallery in central London in September last year.

    James Love, 54, who hid the work after another man had smashed through the door of the gallery, wanted the Banksy “on his walls”, jurors were told.

    The limited edition print was recovered by the Met Police when Mr Love told them where to find it.

    Kingston Crown Court heard the other man, 48-year-old Larry Fraser, had already pleaded guilty to burglary.

    Prosecution barrister Philip Stott said Mr Love was a “relatively successful builder” whereas Fraser was “impoverished” and only had £1.75 in his bank account the day before the theft.

    Mr Love had transferred £200 to Fraser on the day of the offence, Mr Stott said.

    On the evening the Banksy was taken, the mobile phones of both men were being used in the vicinity of the gallery, the jury was told, and Fraser is said to have called Mr Love minutes after smashing the gallery doors and taking the painting.

    After leaving the gallery, Fraser took the print into a nearby property where he left the artwork and changed his top, the trial heard.

    Soon after, the night watchman at the building was “startled” to find a “large painting” by an interior doorway and had “no idea how it had got there because it hadn’t been there when he walked past the same spot earlier that evening”, Mr Stott said.

    He claimed the duo pushed past the night watchman, collected the artwork and drove to a block of flats in the Isle of Dogs where Mr Love unloaded the Banksy.

    Mr Stott said there was a “relatively large number of pictures, about four, featuring love hearts” on the walls of Mr Love’s home and that the stolen work was “consistent with those others”.

    “Given that it was Mr Love who had multiple pieces of art based on love hearts already on his walls, it was likely stolen with a view to it eventually, when the heat had died down, ending up in the same place.”

    Mr Love, of North Stifford, Grays, Essex denies burglary, and the trial continues.

    ISLAMABAD, Sep 15 (APP):Islamabad and Kazakhstan’s capital Astana are moving towards stronger city-level cooperation, with proposals for cultural exchanges, eco-friendly projects, and the symbolic naming of major roads after national icons of both countries.

    The development came during a meeting between Kazakhstan’s Ambassador, Yerzhan Kistafin, and Chairman Capital Development Authority (CDA) and Chief Commissioner Islamabad, Muhammad Ali Randhawa, at CDA Headquarters on Monday.

    Chairman Randhawa emphasized the need to formalize Sister City relations between Islamabad and Astana, noting that enhanced collaboration could boost cultural, environmental, and tourism links.

    “Cooperation between Islamabad and Astana at the city level should be enhanced so that Sister City relations can be formally established,” he said.

    Ambassador Kistafin praised CDA’s ongoing efforts to improve and beautify Islamabad, and proposed a significant cultural gesture: naming a major road in Astana after Quaid-e-Azam Muhammad Ali Jinnah as a symbol of friendship.

    In return, he suggested Islamabad name a prominent road after Kazakhstan’s renowned poet Abai Qunanbaiuly, further cementing literary and cultural ties.

    Randhawa also briefed the ambassador on Islamabad’s eco-friendly initiative, the “Gardenia Hub” project, describing it as a model nursery that would “not only help make the city green but also nurture a spirit of love for plantation among citizens.”

    He added that development and beautification work in the Diplomatic Enclave was underway, including upgraded sports and recreational facilities for diplomats.

    At the conclusion of the meeting, both sides expressed a shared resolve to strengthen cooperation in culture, environment, and urban development, with a focus on long-term city-to-city partnerships.

    Afghanistan Cricket Board (ACB) on Monday replaced right-arm pacer Naveen-ul-Haq with pacer Abdullah Ahmadzai amid the ongoing ACC Men’s T20 Asia Cup 2025 due to a shoulder injury.

    In a statement, the cricket board cited his injury as the cause of dropping out, saying that he was recovering from a shoulder injury, he was not declared fit by the medical team, so he would not play the remaining matches of the tournament.

    “Afghanistan’s veteran fast bowler, Naveen Ul Haq, has been ruled out of the ACC Men’s T20 Asia Cup 2025,” the ACB said in a statement.

    “He is still recovering from a shoulder injury and has not been declared fit by the ACB medical team to participate in the remaining matches. Naveen will continue to undergo intensive treatment and rehabilitation until he is fully fit,” it added.

    Naveen has been replaced by emerging pacer Abdullah Ahmadzai, who made his T20I debut against the United Arab Emirates (UAE) in the recently held T20I tri-series.

    “Fast bowler Abdullah Ahmadzai, who was previously in the reserves and recently made his international debut, has been promoted to the main squad for the Asia Cup 2025,” the ACB’s statement continued.

    “The ACB wishes Naveen Ul Haq a speedy recovery and extends its best wishes to Abdullah Ahmadzai for the matches ahead.”

    For the unversed, Afghanistan has thus far played only one match in the Asia Cup 2025 – the curtain raiser in Dubai – where they beat Hong Kong by 94 runs. Their next group-stage match is scheduled to be played at Zayed Cricket Stadium in Abu Dhabi on Tuesday.

    FBI warns about two campaigns targeting Salesforce instances

    The FBI on Friday released an alert warning that two hacker groups have been targeting Salesforce instances for extortion and data theft. 

    The groups, tracked as UNC6040 and UNC6395, have been identified in recent attacks using different methods of gaining initial access, according to the FBI. The alert includes indicators of compromise and additional guidance that can help security teams determine if they have been targeted and prevent future attacks. 

    UNC6040 has used voice phishing in order to gain access to the Salesforce accounts of targeted organizations. Since October 2024, the group has used social-engineering tactics to get customer-support agents to hand over employee credentials, according to the alert.

    Salesforce previously warned about these social engineering attacks in March, and researchers from Google Threat Intelligence Group warned about UNC6040 in June. 

    The FBI said some victims have received extortion demands from hackers calling themselves ShinyHunters. The demands have arrived between several days and several months after the breaches. 

    The attacks from UNC6395 relied on compromised OAuth tokens for Salesloft Drift, an AI-based chatbot that was integrated with Salesforce. The hackers in these attacks stole data after compromising victims’ Salesforce instances.

    By late August, the companies had revoked all active access and refreshed their tokens, preventing the hackers from further accessing Salesforce platforms through Salesloft Drift, according to the FBI. 

    The supply chain attacks potentially affected hundreds of organizations, and multiple security companies disclosed potential breaches of their customers. 

    Security researchers told Cybersecurity Dive that a threat group claiming ties to ShinyHunters, Scattered Spider and Lapsus$ — which claimed responsibility for hacking Jaguar Land Rover and other high-profile targets — has taken down its Dark Web sites in recent days. 

    The company has not publicly discussed how hackers gained access to its systems, but it did previously confirm that it was investigating the threat claims. 

    Researchers suspect that move is related to increased law-enforcement activity, but it remains unclear if authorities have arrested any of the hackers. Experts also said the attackers are likely to rebrand under a different name rather than completely cease their activities.

    It is not clear whether the attack on JLR is related to the activity described in the FBI alert. The bureau did not respond to a request for comment.

    Cannabis use may quadruple the risk of developing diabetes

    Cannabis use is linked to an almost quadrupling in the risk of developing diabetes, according to an analysis of real-world data from over 4 million adults, being presented at this year’s Annual Meeting of The European Association for the Study of Diabetes (EASD) in Vienna, Austria (15-19 Sept).

    Cannabis use is increasing globally with an estimated 219 million users (4.3% of the global adult population) in 2021, but its long-term metabolic effects remain unknown. While some studies have suggested potential anti-inflammatory or weight management properties, others have raised concerns regarding glucose metabolism and insulin resistance, and the magnitude of the risk for developing diabetes hasn’t been clear.

    To strengthen the evidence base, Dr. Ibrahim Kamel from the Boston Medical Center, Massachusetts, USA and colleagues analysed electronic health records from 54 healthcare organisations (TriNetX Research Network, with centres from across USA and Europe) to identify 96,795 outpatients (aged between 18 and 50 years, 52.5% female) with cannabis-related diagnoses (ranging from occasional use to dependence, including cases of intoxication and withdrawal) between 2010 and 2018.

    They were matched with 4,160,998 healthy individuals (with no record of substance use or major chronic conditions) based on age, sex, and underlying illnesses at the start of the study, and followed for 5 years.

    After controlling HDL and LDL cholesterol, uncontrolled high blood pressure, atherosclerotic cardiovascular disease, cocaine use, alcohol use and several other lifestyle risk factors, the researchers found that new cases of diabetes were significantly higher in the cannabis group (1,937; 2.2%) compared to the healthy group (518; 0.6%), with statistical analysis showing cannabis users at nearly four times the risk of developing diabetes compared to non-users.

    While the authors note that more research is needed to fully explain the association between cannabis and diabetes, it may come down to insulin resistance and unhealthy dietary behaviours. Nevertheless, the study’s results have immediate implications for metabolic monitoring practices and public health messaging.

    As cannabis becomes more widely available and socially accepted, and legalized in various jurisdictions, it is essential to understand its potential health risks. These new sights from reliable real-world evidence highlight the importance of integrating diabetes risk awareness into substance use disorder treatment and counselling, as well as the need for healthcare professional to routinely talk to patients about cannabis use so that they can understand their overall diabetes risk and potential need for metabolic monitoring.”

    The authors note that more research is needed on the long-term endocrine effects of cannabis use and whether diabetes risks are limited to inhaled products or other forms of cannabis such as edibles.

    Despite the important findings, this is a retrospective study and cannot prove that cannabis use causes diabetes, and the authors cannot rule out the possibility that other unmeasured factors may have influenced the results despite efforts to reduce confounding bias via propensity score matching. This study has limitations due to lack of detailed cannabis consumption data and potential misclassification. The authors acknowledge inherent limitations of real-world data often result from inconsistent patient reporting in electronic medical records. They also note that there is a risk of bias because of imprecise measures of cannabis exposure and the reliance on participants to accurately report any cannabis use, even when they lived in places where the drug is illegal.

    Nick Cave has announced a UK exclusive headline show in Brighton

    Singer Nick Cave has announced a UK exclusive headline show in East Sussex next year.

    The Australian musician said he would play “a massive gig”, along with the Bad Seeds, in Preston Park, Brighton, on 31 July 2026.

    “I am thrilled beyond words to return to my beloved Brighton. It’s a homecoming. It’s going to be big, bad and beautiful,” he said.

    Cave, known for songs including Red Right Hand and Into My Arms, added that he was trying to get other musicians to join the show.

    ‘Very special place’

    The 67-year-old lived in Brighton for more than two decades, before moving to Los Angeles following the death of his son, Arthur, in 2015.

    He said he has a “unique and enduring” connection to the city, which has inspired many of his songs, lyrics and films.

    “It’s a very special place to me,” he said.

    “It still has huge significance and meaning.”

    However, said that he did not like the city when he first visited and his wife “dragged” him to live there.

    “I always caught the wrong day or something. It just seemed kind of bleak and depressing,” he told BBC Radio Six Music.

    “But then I came and lived [in Brighton] and it was anything but that,” he added.

    “So much has happened here. The good and the bad.”

    Cave is touring Australia and New Zealand in the New Year as part of the Wild God Tour.
