    Stem Cells of Secondary Progressive Multiple Sclerosis Drive Increased Proinflammatory T-Cell Activity

    A newly published single-center observational study in Neurology Neuroimmunology & Neuroinflammation reported that mesenchymal stem cells (MSCs) from patients with secondary progressive multiple sclerosis (SPMS) may promote inflammatory T-cell responses, contrasting with MSCs from healthy controls. The study authors also identified potential targets to reduce this effect.1 

    The study included 5 patients with SPMS and 7 healthy controls, from whom MSCs were obtained via bone marrow aspiration. Investigators tested the effects of these MSCs on autologous peripheral blood mononuclear cells (PBMCs), focusing on T-cell expression of IL-17, IFN-γ, and GM-CSF. All told, MSCs from healthy individuals reduced the proportion of T cells expressing IL-17 (Th17, P = .046), IFN-γ (Th1, P = .03), and GM-CSF (P = .012). By contrast, MSCs from patients with SPMS increased Th17 cells (P = .01) and GM-CSF–expressing T cells (P = .03), with no significant changes observed in Th1 cell abundance.

    “The lack of the immunomodulatory potential in MS MSCs that is traditionally attributed to MSCs may be explained by an intrinsic deficit in immune regulation, as supported by the evidence of a numeric and/or functional Treg deficit and by the coexistence with other autoimmune disorders,” lead author Radu Tanasescu, MD, PhD, consultant neurologist at Nottingham University Hospitals NHS Trust, and colleagues wrote. “Alternatively, long exposure to an inflammatory milieu may cause the MS MSCs to lose immunomodulatory properties and become proinflammatory.”

    Tanasescu et al continued, “This has implications for autologous MSC treatment of autoimmune diseases including MS. Possibly, the positive effects of MSCs on inflammation and tissue repair are counteracted or diminished by these paradoxical proinflammatory effects. This may explain the negative or only modest results of clinical trials of MSCs in MS. This study, however, also identifies some possible ways of abrogating these proinflammatory effects.”

    Further analysis showed that unstimulated MSCs from patients with SPMS produced lower levels of the anti-inflammatory cytokine IL-10 (P = .03) and higher levels of osteopontin (OPN, P = .002) compared with MSCs from healthy controls. Gene expression profiling identified increased expression of ADAM-28 in MSCs from patients with SPMS, which was subsequently confirmed at the mRNA level by quantitative polymerase chain reaction (qPCR) and at the protein level by flow cytometry.

    The study also explored strategies to mitigate the proinflammatory effects of MS-derived MSCs. When MSCs from patients with SPMS were co-cultured with autologous PBMCs in the presence of natalizumab (Tysabri; Biogen), a monoclonal antibody that binds α4β1 and blocks its interaction with ADAM-28 and OPN, the proportion of Th17 cells decreased. Similarly, supplementing the co-cultures with IL-10 prevented the increase in Th17 cells observed in the absence of intervention. Overall, these findings suggested that blocking ADAM-28 and OPN interactions or increasing IL-10 levels can reduce the proinflammatory potential of MSCs from patients with SPMS.

    All told, the study included a small number of donors, and limited PBMC availability restricted the number of experiments. Authors also noted that minor age differences existed between patients with SPMS and healthy controls, which could influence MSC or immune aging. However, researchers expressed that these differences were not statistically significant and there were no patients classified as older as defined for MSC purposes.2 The study did not examine effects on regulatory T cells because of cell limitations. In addition, molecules for targeted analysis were chosen based on functional pathway analysis rather than unbiased differential gene expression, though authors noted that multiple assays supported their relevance.

    “In conclusion, we provide evidence for a proinflammatory effect of MSCs from [patients with] MS on autologous immune cells and further show that targeting some molecules that are differentially expressed between [patients with] MS and non-MS controls can counteract this proinflammatory effect. These observations can lead to the development of modalities for improving outcomes of MSC treatment of MS,” Tanasescu and colleagues noted.1

    Kate Middleton and King Charles III shared a royally sweet moment during an emotional event.

    After attending the Sept. 16 funeral for Katharine, Duchess of Kent—the wife of Queen Elizabeth II‘s cousin Prince Edward, Duke of Kent, the Princess of Wales was photographed putting an arm on her father-in-law’s shoulder and keeping it there while kissing him on both cheeks and curtsying.

    In response to the touching gesture, the 76-year-old monarch looked down for a moment before peering back at Kate, with his arm placed gently by her side.

    As for Prince William, he looked somber while standing by the princess and his father during the funeral proceedings.

    Meanwhile, Charles’ wife Queen Camilla was unable to attend due to illness, with Buckingham Palace saying in a statement that she “is recovering from acute sinusitis,” or a sinus infection.

    Charles and Kate, 43, have also been going through challenging health battles as of late: Both were diagnosed with cancer in 2024.

    The King—who, like Kate, has not detailed his cancer diagnosis—last shared an update on his health earlier this month.

    Per Hello!, Charles told a fellow cancer patient, “I’m not too bad” after being asked about his recovery during a royal visit to the Midland Metropolitan University Hospital.

    “Half the problem is detecting it, isn’t it, in time,” the patient, Matthew Shinda, 73, quoted the King as saying. “The great thing I think is they’re getting better and better at dealing with these things.”

    Charles previously reportedly shared an update about his health with another fan during a visit to Jockey Club Rooms in Newmarket in July, noting that he was in a good place.

    After the princess—who is in remission—revealed her diagnosis in March of that year, the King’s rep noted to NBC News that Charles has “remained in the closest contact with his beloved daughter-in-law throughout the past weeks” and that he and Camilla “will continue to offer their love and support to the whole family through this difficult time.”

    Charles—who, like Kate, has not detailed what type of cancer he was diagnosed with—last shared an update on his health earlier this month.

    “You put on a sort of brave face, stoicism through treatment,” she said during a visit to Colchester Hospital in Essex, per People. “Treatment’s done, then it’s like, ‘I can crack on, get back to normal,’ but actually, the phase afterwards is really, really difficult.”

    Daily weight loss pill can help cut body weight by a fifth, trial shows | Obesity

    A daily pill for weight loss can help people reduce their body weight by as much as a fifth, according to a trial that could pave the way for millions more people to shed pounds.

    The drug, called orforglipron, is manufactured by Eli Lilly and targets the same GLP-1 receptors as weight loss injections such as Mounjaro and Wegovy. In a trial of 3,127 adults, one in five people who took the once-a-day tablet for 72 weeks lost 20% or more of their body weight.

    Weight loss jabs have been transformative but pill versions are seen as a holy grail because they are easier to store, distribute and administer and are also expected to be cheaper, offering fresh hope for the millions of people trying to lose weight.

    Orforglipron is a GLP-1 agonist, a type of medication that helps lower blood sugar levels, slows the digestion of food and can reduce appetite.

    The weight loss seen among people taking the tablet is not as stark as that among patients taking tirzepatide (Mounjaro), which is also made by Eli Lilly, but experts believe the tablet will be more accessible and convenient compared with injections.

    Orforglipron is not yet approved by the US Food and Drug Administration (FDA) or regulators in other countries. Eli Lilly has said it expects substantial demand when the new pill is launched.

    The company published a snapshot of the results in August and the full paper detailing the findings has now been published in the New England Journal of Medicine and presented to the annual meeting of the European Association for the Study of Diabetes in Vienna, Austria.

    In the study, the 3,127 patients were split into groups taking orforglipron pills of different strengths, while others took a placebo, for 72 weeks.

    All of the patients had obesity, meaning they had a body mass index (BMI) score of 30 or over, but they did not have diabetes. Patients from the US, China, Brazil, India, Japan, South Korea, Spain, Slovakia and Taiwan took part in the study.

    Researchers led by Dr Sean Wharton, of McMaster University in Canada, found that after 72 weeks people taking the lowest dose of orforglipron, 6mg daily tablets, lost an average of 7.5% of their body weight. Those taking the highest dose, 36mg, lost an average of 11.2% of their body weight.

    Among patients taking the highest doses, 54.6% had a reduction of 10% or more of body weight, 36% had a reduction of 15% or more, and 18.4% had a reduction of 20% or more.

    Researchers said other health metrics also improved among people taking the drug, including better blood pressure, a smaller waist circumference and a reduction in bad cholesterol levels.

    The most common side-effects were gastrointestinal, which were said to be mild to moderate.

    “In adults with obesity, 72-week treatment with orforglipron led to significantly greater reductions in body weight than placebo,” the authors wrote. “The adverse-event profile was consistent with that of other GLP-1 receptor agonists.”

    Wharton said: “This could mean an expansion of obesity interventions to groups who are currently excluded due to the cost of and lack of access to injectable medications.”

    Health leaders have hailed weight loss drugs as transformative. But injections come with additional work for health services, so tablet forms could help millions more people trying to lose weight.

    Separate research published in JAMA Pediatrics on Monday shows weight loss jabs could be effective on children as young as six who are obese or have type 2 diabetes. However, stomach problems were “significantly more common” among children taking the drugs, the researchers said.

    They said longer follow-up periods in future trials and more real-world studies were “essential to establish the long-term effects of GLP-1 RAs in children and adolescents”.

    production of the new BMW iX3 will get underway in late October at the

    Debrecen. For example, Plant Debrecen was planned and built digitally
    from the outset, and celebrated its virtual start of production back
    in March 2023 in the BMW Group’s Virtual Factory. This approach
    allowed every operation to be tested virtually in advance, enabling
    production lines to be installed in the buildings exactly as in the
    digital twin.

    The body shop, in particular, benefits from prior
    digital planning and validation. Every process detail was digitally
    simulated in advance to determine the ideal value stream and ensure
    that each of the nearly 1,000 robots was optimally positioned. From
    the outset, close collaboration between development and production
    teams led to high production efficiency and maximum customer benefit.
    For example, the number of joining methods was significantly reduced,
    lowering overall complexity. Design features integrated early in
    development are also evident in the details of the Neue Klasse, such
    as the invisible door seal. This creates a visual connection between
    the window and the door, resulting in a unique look. In addition,
    maximising the space available for the battery in the body structure –
    and thus the size and performance of the battery – directly benefits
    the customer.

    At BMW Group Plant Debrecen, the paint shop plays a
    critical role in significantly reducing the CO2e
    footprint of the BMW iX3
    . Production of the new BMW iX3
    will generate a total of about 80kg CO₂e (scope 1/2 emissions). This
    figure covers CO₂e emissions from Plant Debrecen, as well as in-house
    parts production at other BMW Group facilities, including components
    manufactured in Landshut, for example. This represents a
    reduction of approx. two-thirds compared to production of
    existing BMW derivatives.
    For Plant Debrecen alone, this
    approach will reduce CO₂e emissions from the manufacture of a
    vehicle, including its high-voltage battery, by around 90%

    – to about 34 kg CO₂e (when operating at full capacity, compared with
    other BMW Group facilities).

    Paint shops typically rely on gas to reach the high temperatures (up
    to 180 degrees Celsius) required. Plant Debrecen will be the BMW
    Group’s first car plant supplied exclusively with electricity
    from renewable energy sources during normal operation
    , i.e.
    without using fossil fuels such as oil or gas. Because of its high
    energy demands, the paint shop is the most important factor in
    significantly lowering Plant Debrecen’s CO2e
    emissions. In the paint shop alone, using electricity from
    renewable sources reduces annual CO2e emissions by up to
    12,000 tonnes.
    About a quarter of the plant’s annual power
    needs will be supplied by the 50‑hectare on-site photovoltaic
    system.
    Surplus solar energy, such as that generated on
    non‑working days, is stored in a 1,800 m³ thermal storage
    system
    with a capacity of 130 MWh.

    The paint shop also uses an energy recovery system. A heat
    grid
    was successfully implemented in the design of the new
    paint shop, delivering additional energy savings of up to 10%. This
    innovative concept combines multiple measures for efficient
    energy recovery from the compressed air supply,
    drying ovens and cooling systems. The recovered waste heat is then
    used to preheat the water circuit.

    In addition, a thermal storage system – holding
    1,800 m³ of water, with a capacity of 130 MWh – stores surplus energy
    from the photovoltaic system during off-peak periods and supplies it
    as heat during peak demand.

    Full digitalisation of production processes further boosts
    assembly efficiency. The AIQX (Artificial
    Intelligence Quality Next) IT platform developed in-house at the BMW
    Group is a key component of the BMW iFACTORY. AIQX uses sensors and
    camera systems along the production line to automate quality
    processes. AI evaluates the data and provides real-time feedback to
    employees on the line. Looking ahead, vehicles on the line will also
    become active, connected participants in the industrial IoT (Internet
    of Things) ecosystem. They will conduct self-analysis, interact with
    plant employees in real time and automatically share and document
    relevant messages, using tools such as on-board cameras and IT
    systems. The equipment, tools, components and every BMW in the
    assembly halls are already digitally connected to the BMW production system.

    The building’s “finger structure” – an optimised version of the
    design employed at BMW Group Plant Leipzig – allows 80% of parts to be
    delivered directly to the correct point of assembly on the line.
    All-electric in-house logistics also benefit from
    autonomous tugger trains, which transport high-voltage batteries
    directly from production to installation points, as well as Smart
    Transport Robots that autonomously deliver smaller components to the
    assembly line. By linking all internal and external databases, the
    entire logistics process achieves unprecedented digital depth,
    automating many previously manual, interconnected analysis tasks.
    Well-structured information is therefore available at any time and can
    be analysed instantly “at the push of a button”.

    The BMW Group has created highly intelligent production processes for
    its new high-voltage batteries developed in-house.
    Plant Debrecen will be the first of five plants worldwide to begin
    series production of Gen6 high-voltage batteries. The production
    processes were initially developed and tested at pilot plants.
    Artificial intelligence, data analytics and constant knowledge sharing
    within the production network also play a key role in the ramp-up of
    production. Digital twins of production and comprehensive AI databases
    ensure optimisation of processes and employee training. A consistent
    zero-defect approach enables seamless in-line quality inspections and
    100% end‑of-line monitoring. In line keeping with the “local for
    local” principle, high‑voltage battery assembly takes place directly
    on site, allowing the production teams to take advantage of
    infrastructure efficiencies and short distances.

    BMW Group Plant Debrecen is the first production site in the
    company’s network not tied to a specific primary plant. Instead, it
    serves as a network plant, combining best practices from various
    locations worldwide. A major advantage of this approach is that the
    plant’s current workforce of more than 2,000
    employees can be trained within the network, allowing
    employee know-how from different locations, such as China, South
    Africa, Mexico, the US and, of course, Germany, to be shared. At the
    same time, employees throughout the global network gain the
    opportunity to share their technology insights in Debrecen and bring
    expertise in production of the Neue Klasse models back to their own
    location – creating a win-win situation for all the plants.

    Earth Has Another Quasi-Satellite: The Asteroid Arjuna 2025 PN7

    Whenever astronomers detect something new moving through our region of space, like an interstellar object or an unusual asteroid, somebody somewhere claims it could be an alien interstellar space probe. It’s like one of those laws about human behaviour—Godwin’s Law for example—that should probably have its own name.

    This applies to the detection of 1991 VG, an asteroid with an Earth-like orbit discovered by the Spacewatch Project in 1991. Astronomers now know that it’s just an asteroid, and they’ve also found others like it. Together, they’re called the Arjuna asteroids and they’re Near-Earth Objects (NEO). There’s more than 100 of them and they constitute the so-called Arjuna secondary asteroid belt.

    Now the Arjuna group has one more member: Arjuna 2025 PN7. It’s discovery is presented in a new research note in Research Notes of the American Astronomical Society titled “Meet Arjuna 2025 PN7, the Newest Quasi-satellite of Earth.” The authors are Carlos de la Fuente Marcos and Raúl de la Fuente Marcos, both from the Universidad Complutense de Madrid, Ciudad Universitaria.

    When 1991 VG was discovered, it followed a path very similar to Earth’s and remained close to it. That’s what led to the ‘alien space probe’ thinking. But it was really just the first member of a newly discovered class of objects. That only became apparent when more were discovered, and now the natural explanation has eliminated the speculation.

    The Arjuna asteroids are known as quasi-satellites. That’s because while they move in resonance with Earth, they’re not gravitationally bound. Sometimes one of the Arjuna asteroids is captured by the Earth for a brief period of time and becomes a mini-Moon. “The Arjunas with the most Earth-like orbits can experience temporary captures as mini-moons of our planet,” the authors write. That happened with the Arjuna asteroid 2024 PT5, which was captured by Earth and posed a small threat of impact for a while.

    “Quasi-satellites are in a resonant orbit but are not gravitationally bound to Earth, allowing for more sustained, though unbound, proximity; while mini-moons are characterized by temporary gravitational captures by Earth, meaning they are gravitationally bound, albeit for a limited time,” the authors explain in their research note.

    Other current quasi-satellites include 164207 Cardea (2004 GU9), 469219 Kamo‘oalewa (2016 HO3), 277810 (2006 FV35), 2013 LX28, 2014 OL339, and 2023 FW13. 2025 PN7 is now the newest member of that group.

    2025 PN7 is also an Apollo asteroid, a larger group of asteroids of which the Arjuna’s are a small subclass. A critical distinction between them is that Arjunas don’t have Earth-crossing orbits while the Apollos do.

    The researchers say that 2025 PN7 won’t be a quasi-satellite for very long. “The available data indicate that 2025 PN7 is a relatively short-lived quasi-satellite of Earth,” they write.

    It’ll only be a quasi-satellite for about 128 years. Eventually, subtle changes in orbital elements will mean it leaves its resonance with Earth and will like follow a Trojan orbit or horseshoe orbit.

    Meta Connect 2025: What to expect and how to watch

    Meta Connect 2025 — the company’s biggest conference of the year where it unveils smart glasses and VR headsets — kicks off on Wednesday night. We’re expecting the star of Meta Connect 2025 to be the company’s new AI-powered smart glasses with Ray-Ban and Oakley, but the company may have some other surprises in store regarding the Metaverse, Quest headsets, or even its broader AI ambitions.

    Meta says it has sold millions of Ray-Ban Meta smart glasses, and earlier this year, Meta unveiled its latest AI-powered smart glasses with Oakley, which were designed for athletes. Silicon Valley is leaning heavily into AI wearables, and Meta seems to be one of the companies leading the charge.

    Notably, this is the company’s first Connect conference since it started Meta Superintelligence Labs (MSL), its boldest effort yet to develop cutting-edge AI systems under former Scale AI CEO Alexandr Wang. It’s possible we’ll get some official updates on how that project is going, and we may hear from some MSL executives.

    As Meta looks to regain its footing in the AI race, this year’s Connect feels especially consequential.

    How to watch

    Meta Connect 2025 starts at 5 p.m. PT Wednesday, with a keynote from CEO Mark Zuckerberg. The event will take place in person at Meta’s headquarters in Menlo Park. You can register for free to watch the livestream virtually on Meta’s website. Meta’s agenda says the keynote will be roughly an hour.

    If you want to get that Menlo Park feel from the comfort of your living room, you can also access the keynote through Horizon via your Meta Quest headset. You can also access the Meta Connect 2025 keynote on Facebook via the company’s official developer page, Meta for Developers.

    On Thursday, Meta will host a Developer Keynote staring 10 a.m. PT to discuss the new experiences people can build with its devices. Then, at 10:45 a.m. PT, Meta will host a conversation between Chief Scientist of Reality Labs Michael Abrash and VP of Reality Labs Research Richard Newcombe. The two Meta executives are slated to discuss the “future of glasses with contextual AI, and how Meta is poised to transform the future of computing.”

    What to expect

    There have already been several leaks regarding what will be announced at Meta Connect 2025. Perhaps the biggest pertains to a new type of smart glasses called Hypernova.

    A now-removed video on Meta’s YouTube channel, spotted by UploadVR, showed a pair of Ray-Ban Meta smart glasses with a heads-up display on the right lens, as well as cameras, microphones, and an onboard AI assistant.

    The glasses in the video were controlled by a wristband, which was unveiled at last year’s Connect and is controlled by subtle hand gestures.

    The video suggests that Meta will unveil, and perhaps launch, the Hypernova glasses this week. CNBC previously reported that Meta was planning to unveil Hypernova and launch the wristband at Connect 2025.

    Meta also seems likely to unveil new pair of smart glasses it developed with Oakley at Connect 2025. The companies are expected to launch a new pair of AI-powered smart glasses in Oakley’s Spheara style. This features a large unified lens on the front — an ideal shape for runners and bikers. Unlike previous Meta smart glasses, this model has one centered camera above the nose bridge, rather than two cameras on the top corners of the frames.

    On the VR front, it’s unclear whether Meta will release any new Quest headsets as part of this year’s Connect. Even though the conference, and company, is named after the Metaverse, it seems like that’s less of the focus this year. Meta is reportedly developing an ultralight VR headset for launch by the end of 2026. However, the company could wait to show that off at next year’s Connect.

    That said, Meta promises that Zuckerberg will talk about the Metaverse in some shape or form. We don’t doubt that.

    As for Meta’s AI ambitions, it wouldn’t be surprising if Zuckerberg used Connect 2025 as a chance to highlight all the work MSL is doing. The company’s first LlamaCon, its AI developer conference, took place earlier this year before Meta invested billions in Scale AI and hired researchers from around the industry.

    Right now, Meta’s standalone AI app is in a confusing spot where it both controls smart glasses and can be used as an AI chatbot. It’s possible that app also gets some updates that make it easier to use.
